Video post can use any key colour (such as blue 0,0,255) although the colour allows for now variation, but for best results you'd need a grayscale mask with anti-aliased edges. I often use animator pro for this. Make the background the key colour, clip the whole animation over time, make pure white the current colour, go into anim cel, do a one colour operation using white and paste every frame. Then I'd go through the whole animation and either soften or unzag the edges. You can use the macro recorder to automate most of the repetitive operations. This resulting flic would then be your alpha channel and used as a rgb mask in video post. If you don't have animator pro maybe hiresQFX 2.0 has a way of automating this.
