From my experience, we generally overprint the black layer…you will have
registration problems if you cutout the letter's shape from the blue (or
yellow) background, unless you trap your letter by stroking it. It is
difficult for most presses to hold perfect registration…the darker your
background color, the more noticable the white (or whatever your paper col
color is) line will be when the letter doesn't land directly in your
knockout. If your letter is, say, yellow on a blue background, you have to
knockout your letter since the color (yellow on blue) will dramatically
change the color of your letter….
